I didn't read careful enough. I thought you were starting the broker in REMOTE mode (i.e. not managed by Glassfish).
Maybe you can try to supply the property through the admin console. Go to Configuration -> Java Message Service. Then there is a box on the right where you can enter Start Arguments. Try putting "-Dimq.cluster.url=file\://opt/glassfish/config/cluster.config" there (without quotes).
